Try a different USB Port.

I had the same problem. I did a soft reset (disconnect Zune from computer, hold back button + up until screen goes black) then fired up the Zune software, and plugged the Zune back in and it worked. I noticed that this happens when I am listening/watching the Zune when I plug it in and then fire up the software.

My machine deteriorated to where I couldn't get the Zune to connect on any of my USB ports. It would launch the Zune software but then go blank and just sit there unsatisfied.
After trying all of the remedies I found on the boards, I started the machine in Safe Mode and found four USB controllers in Hardware Manager that were Zune controller all showing errors. These did not show when I wasn;t in Safe mode. I uninstalled all four of these and things started working the next time I booted and connected my Zune.
