I know of no way to do it, and I don't think it's necessary. I think with the HDD Hack you might be able to do it using System defrag on XP, but I wouldn't... It might mess with your Zune enough to make it so you'd have to restore it an lose your music.

Assuming you could actually get the windows(or 3rd party) defragmenter to see the zune's HDD, its probably best Not to attempt defragmentation on the Zune. It would be pushing the zunes HDD to the max in terms of read/write speed and might screw up the filesystem. There is no reason to defragment the Zune anyway.
